: The HIR2 monoclonal antibody reacts with human glycophorin A, a sialoglycoprotein expressed by erythroid precursors and mature circulating red cells. Applications Reported: HIR2 can be used in flow cytometric analysis. Applications Tested: The HIR2 (also GA-R2) antibody has been tested by flow cytometric analysis of normal human peripheral blood cells and cell agglutination assay and can be used at approximately 0.1 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 μm post-manufacturing filtered.Glycophorins A (GPA) and B (GPB) which are single pass membrane sialoglycoproteins. GPA is the carrier of blood group M and N specificities, while GPB accounts for S and U specificities. GPA and GPB provide the cells with a large mucin like surface and it has been suggested this provides a barrier to cell fusion, so minimizing aggregation between red blood cells in the circulation.
