The B-type cyclins serve as subunits of the major mitotic protein kinase in many different organisms. In mammals, Cyclin B is a subunit for the cdc2 (Cdk1) protein kinase and is synthesized during late S and G2 phases. Cyclin B associates with inactive cdc2 and facilitates phosphorylation of cdc2 at Thr-14 and Tyr-15. This phosphorylation maintains the inactive state until the end of G2. The inactive Cyclin B-cdc2 complex continues to accumulate in the cytoplasm until the CDC25 phosphatase dephosphorylates Thr-14 and Tyr-15, rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when Cyclin B becomes degraded. This degradation process appears to be ubiquitin-dependent and is necessary for the cell to exit mitosis. Also, the conserved domain in B-type cyclins, known as the P box, is required for CDC25 activation. This antibody is routinely tested by western blot analysis. Other applications were tested at BD Biosciences Pharmingen during antibody development only or reported in the literature.
