Details

The Human Soluble CD105/Endoglin (Hu sCD105) ELISA quantitates Hu sCD105 in human serum, plasma, buffered solution, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu sCD105. Principle of the method The Human sCD105 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. See manual for more information on validation.CD105 (Endoglin is a Type I transmembrane protein) is highly expressed on human vascular endothelial cells and exists on an O- and N-glycosylated homodimer. Up regulation of CD105 expression has been demonstrated in tumor vasculature and proliferating cells, suggesting that it is a proliferation associated endothelial marker. CD105 is highly expressed on endothelial cells and promotes angiogenesis during wound healing, infarcts and in a wide range of tumors and its gene expression is stimulated by hypoxia. CD105 binds to TGF beta 1 and 3 with high affinity but not to TGF beta 2. Mutations in the CD105 gene cause hereditary hemorrhagic telangiectasia, also known as Osler-Rendu-Weber syndrome 1, an autosomal dominant multisystemic vascular dysplasia. Alternatively spliced transcript variants encoding different isoforms of CD105 have been found. Diseases associated with CD105 dysfunction include hereditary hemorrhagic telangiectasia.
