The Human soluble CD134/OX40 ELISA quantitates Hu CD134/OX40 in human serum, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu CD134/OX40. Principle of the method The Human CD134/OX40 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.OX40 (CD134) is a member of the TNF-receptor superfamily, and has been shown to activate NF-kappaB through its interaction with adaptor proteins TRAF2 and TRAF5. Knockout studies in mice suggested that OX40 promotes the expression of apoptosis inhibitors BCL2 and BCL2lL1/BCL2-XL, and thus suppresses apoptosis. The knockout studies also suggested the roles of this receptor in CD4 T cell response, as well as in T cell-dependent B cell proliferation and differentiation.OX40 is not constitutively expressed on resting naïve T cells, unlike CD28. OX40 is a secondary costimulatory molecule, expressed after 24 to 72 hours following activation. OX40L, a ligand of OX40, is also not expressed on resting antigen presenting cells. Expression of OX40 is dependent on full activation of the T cell; without CD28, expression of OX40 is delayed and expressed at lower levels.
