The Human Soluble L-Selectin/CD62L (Hu sCD62L) ELISA quantitates Hu sCD62L in human serum, plasma, cell culture supernatants, or amniotic fluid. The assay will exclusively recognize both natural and recombinant Hu sCD62L. Principle of the method The Human sCD62L solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.CD62L (L-selectin) is an adhesion glycoprotein with a molecular weight range of 74-95 kDa that is constitutively expressed on the cell surface of leukocytes, and mediates their homing to inflammatory sites and peripheral lymph nodes by enabling rolling along the venular wall. CD62L is also involved in activation-induced neutrophil aggregation. Activation-dependent CD62L shedding, however, counteracts neutrophil rolling. CD62L has also signaling roles including enhance of chemokine receptor expression. Similar to CD62P, the major ligand of CD62L is PSGL-1 (P-selectin glycoprotein ligand-1). Single-nucleotide polymorphisms in the CD26L gene have been associated with various diseases including immunoglobulin A nephropathy. Alternatively spliced transcript variants have been found for CD26L.
