The Mouse Interleukin-23 (Ms IL-23) ELISA quantitates Ms IL-23 in mouse serum, plasma, buffered solution, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms IL-23. Principle of the method The Mouse IL-23 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.IL-23 is a heterodimeric cytokine composed of the p40 subunit of IL-12 that is disulfide-linked with to two subunits, a p19 subunit unique to IL-23, and a p40 subunit that is shared with IL-12. IL-23 interacts with IL-12Rbeta1 and a beta2-like receptor subunit with STAT4 binding domain, IL-23R. IL-23 is secreted by activated mouse and human dendritic cells and is involved in the inflammation process and associated with autoimmune diseases. Mouse IL-23 was found not to induce significant amounts of IFN-gamma, induces strong proliferation of memory T cells (but not naive T cells), and activate mouse memory T cells to produce the proinflammatory cytokine IL-17. On the other hand, human IL-23 induces proliferation of memory T cells, induces moderate levels of IFN-gamma, and is produced by naive and memory T cells. IL-23 plays an important role in stimulating memory T-cells, and is required for the induction, expansion, maintenance, and downstream effector functions of Th17 cells, which play a vital role in upregulating neutrophil chemokines and various pro-inflammatory cytokines. IL-23 interacts with the receptors IL-12R beta1 and IL-23R to activate the Jak-Stat signaling cascade. IL-23 has been shown to possess potent anti-tumor and anti-metastatic activity in mouse models of cancer, suggesting a potential role for IL-23 in therapeutic treatment of cancer.
