The Mouse Inflammatory Protein 10/CXCL10 (Ms IP-10) ELISA quantitates Ms IP-10 in mouse serum, plasma, buffered solution, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms IP-10. Principle of the method The Mouse IP-10 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Binding of CXCL10 to CXCR3 results in pleiotropic effects, including stimulation of monocytes, natural killer and T-cell migration, and modulation of adhesion molecule expression. CXCL10 is a pro-inflammatory chemoattractant cytokine that attracts activated T lymphocytes. CXCL10 is constitutively expressed at low levels in lymph organs, and the expression is highly induced by IFN- gamma and to some extent by IL12. It is an important biomarker for diagnosis of many clinical conditions, and it plays an important role in host defense and inflammation. CXCL10 exhibits an angiostatic and an antitumor role that is also involved in Th1 inflammatory diseases, Hashimoto's thyroiditis, Graves′ disease and Type 1 diabetes mellitus.
