The Human ICOS (CD278) ELISA quantitates Hu ICOS in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu ICOS. Principle of the method The Human ICOS solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. See manual for more information on validation.CD278 (ICOS) belongs to the CD28 and CTLA-4 cell-surface receptor family, forms homodimers, and is highly expressed in activated T-cells. CD278 is essential both for efficient interaction between T and B-cells and for normal antibody responses to T-cell dependent antigens. CD278 enhances all basic T-cell responses to a foreign antigen, namely proliferation, secretion of lymphokines, up-regulation of molecules that mediate cell-cell interaction, and effective help for antibody secretion by B-cells. Diseases associated with CD278 dysfunction include Common Variable 1 Immunodeficiency and ICOS-related Common Related Immunodeficiency.
