Mouse IFN alpha A Recombinant Protein, 121001, from Invitrogen., 121001, from Invitrogen. Species Reactivity: Mouse; Applications: ELISA, Immunoprecipitation, Western Blot.IFN alpha (Interferon alpha) is a Type I interferon belonging to a family of structurally related, species-specific proteins exclusive to vertebrates. The interaction of IFN-alpha with its receptor component result in the activation of a number of signaling pathways which are mainly involved in innate immune response against viral infection. In humans, the gene for IFN alpha is located on on chromosome 9. The type I interferons consist of 14 different alpha isoforms (subtypes with slightly different specificities), and single beta, omega, epsilon and kappa isoforms. Homologous molecules are found in many species, including rats and mice (and most mammals), and have been identified in birds, reptiles and fish species. IFN alpha is produced primarily by plasmacytoid dendritic cells, is a potent component of the anti-viral innate immune response and modulates adaptive immunity. Diseases associated with the dysfunction of IFN alpha include Hepatitis C and Hepatitis. Assay Used to Measure Bioactivity: Interferon was titrated with the use of the cytopathic effect inhibition assay as described [Familletti, et al. (1981), Methods in Enzymology, Vol. 78 (S. Pestka, ed.), Academic Press, New York, 387-394] with the exception that EMCV rather than VSV was used as the challenge. The activity was determined relative to a lab standard of Mu IFN-αA which was calibrated to the NIH mouse IFN-alpha standard (Ga02-901-511). Mouse (L929/EMCV) in this assay the EC50 for IFN is ∽ 5U/mL. Lot Activity was derived from multiple determinations in the above assay. Store at -70°C or below for retention of full activity. Thaw product vial by incubation in cold tap water until just thawed. Aliquot and refreeze unused contents to minimize effects of freeze thawing. Refreezing should be done on dry ice or in a dry ice/alcohol bath. Further dilution of product should be in buffers containing protein such as 0.1% BSA or tissue culture media with serum. Dilution for freezing of material below 2x10^5 units/mL is not recommended. One freeze thaw cycle is equivalent to thawing an aliquot prepared from the material received.
